Inhibitory effect of Ketotifen on rat isolated uterus contraction in compare with a β2-adrenoceptor agonist and an ATP-dependent K channel activator

Abstract:

کتوتیفن یک بنزوسیکلوهپتاتیوفن با فعالیت های متعدد فارماکولوژیکی از جمله اثر مهاری بر روی انقباضات روده و مثانه است ولی اثرات این دارو بر روی رحم بررسی نشده است. در این تحقیق اثر کتوتیفن بر روی انقباضات ناشی از استیل کولین و کلرید پتاسیم در رحم جدا شده رات (In vitro) توسط دستگاه فیزیوگراف اندازه گیری و با اثر متاپروترنول و دیازاکسید مقایسه گردید. کتوتیفن انقباضات ناشی از کلرید پتاسیم را به صورت وابسته به غلظت مهار کرد (pD2=6.2±0.13: لگاریتم منفی قدرت دارو) و موجب 25 برابر جابجایی منحنی غلظت-پاسخ به سمت راست در غلظت 5µM گردید. اثر کتوتیفن بر روی انقباضات ناشی از کلرید پتاسیم مشابه اثر متاپروترنول است و موجب مهار انقباضات با غلظت های مختلف کلرید پتاسیم و کاهش حداکثر پاسخ ایجاد شده در بافت گردید. این مطالعه نشان می دهد که کتوتیفن یک مهار کننده انقباضات رحم جدا شده است. از آنجا که اساس درمان زایمان های زودرس بر مهار انقباضات زودرس رحم استوار است، در صورتی که کتوتیفن اثرات مشابهی بر روی رحم در In vitro هم داشته باشد این دارو ممکن است برای درمان این عارضه مفید باشد.
